Snooki and pals knock back breast milk shots

Nicole "Snooki" Polizzi and Jenni "JWoww" Farley are no strangers to knocking back a shot (or 10), but on the latest episode of "Snooki & JWoww," the pair hesitated -- and so did Snooki's main man, fianc? Jionni LaValle.

That's because the drink of the day was Snooki's very own breast milk, served warm.

When handed the shot, Snooki cried "Ew!" In fact, at first, the only taker was JWoww's fianc? Roger Mathews, who downed it fast and waited for Jionni to follow suit. When the father of baby Lorenzo refused, JWoww lashed out.

"You just allowed another man to drink your woman's breast milk before you?!" she said with disbelief.

But it was hard to blame Jionni for holding out, especially after Roger delivered his taster's review.

"It's clumpy going down," he mused. "It was like chugging mashed potatoes."

Eventually they all gave it try, but there's one person who couldn't be convinced to join in the fun.

On Wednesday, Snooki and JWoww made an appearance on "Anderson Live," and tried to talk Anderson Cooper into taking a swing of Snooki's offering.

"There's no way! I'm sorry," the clearly-revolted host said. "I really can't. I'm lactose intolerant."

But unlike what the gang was sipping on MTV ("That was her real boob milk," JWoww informed the audience), there was nothing but sweet almond milk in Cooper's cup.

It's not that Snooks was unwilling to share the real deal with him, it just that she's not lactating anymore.

Genetic evidence that new therapies targeting Parkinson's disease may cause harm

Mar. 20, 2013 ? NorthShore University HealthSystem (NorthShore) and Mayo Clinic researchers have partnered on a study that shows genetic and clinical evidence that therapies targeting the expression of alpha-synuclein -- a gene whose function is involved in the development and progression of Parkinson's disease -- may accelerate disease progression and increase the risk of physical incapacitation and dementia. If replicated, the findings will have profound implications for therapies under development for Parkinson's disease.

"Our research suggests therapies that seek to suppress alpha-synuclein in Parkinson's disease may actually accelerate the disease process and increase the risk for developing severe physical disability and dementia," says lead author Demetrius Maraganore, M.D., Ruth Cain Ruggles Chairman, Department of Neurology at NorthShore. "We believe it is our responsibility to release these data because this type of treatment may have long-term harmful effects."

Alpha-synuclein is a major component of Lewy bodies -- a characteristic brain cell abnormality that occurs in all cases of Parkinson's disease. Since its discovery as a cause of familial Parkinson's disease nearly 20 years ago, alpha-synuclein has been the focus of intensive efforts by researchers working to definitively characterize the protein's role in idiopathic Parkinson's disease and its potential as a target for neuroprotective therapies. It has also been the focus of multiple efforts to develop a molecule that suppresses the protein function. A vaccine that targets alpha-synuclein (reducing alpha-synuclein levels) is currently in Phase I clinical trials, and a number of molecules that target the protein for reduction are in advanced stages of preclinical development.

"For the first time we observed that while over-expression of alpha-synuclein increases the risk for developing Parkinson's disease, conversely, under-expression is associated with worse motor and cognitive outcomes after the disease starts," says first author Katerina Markopoulou, M.D., Ph.D., a neurologist at NorthShore. "This raises concerns about the efficacy and safety of therapies designed to reduce alpha-synuclein expression in Parkinson's disease."

The researchers followed 1,098 Mayo Clinic patients for nearly 15 years (median: eight years), and sequenced the patients' DNA to determine the presence of gene variants that regulate how much alpha-synuclein protein is made. They studied the association of these gene variants with patients' survival that was free of severe motor and cognitive disabilities. Patient outcomes were measured by telephone interviews.

The scientists found that patients who had the reduced expression genotype had a 23 percent greater risk of becoming wheelchair-dependent or developing dementia.

"This is the first large genetic association study of alpha-synuclein and longitudinal outcomes in Parkinson's disease," says Eric Ahlskog, M.D., Ph.D., a Mayo Clinic neurologist and author on the study. "If replicated, this research may change the treatment paradigm focused on alpha-synuclein reduction for Parkinson's disease."

The study will be discussed at 5 p.m. EST, March 20 at the 2013 American Academy of Neurology (AAN) Annual Meeting in San Diego. This research is one example of the collaborative efforts between NorthShore and Mayo Clinic under the Mayo Clinic Care Network, a unique partnership that provides NorthShore patients with access to medical resources and experts from both systems working together on their behalf.

Potential immune benefits of strong vitamin D status in healthy individuals

Mar. 20, 2013 ? Research from Boston University School of Medicine (BUSM) shows that improving vitamin D status by increasing its level in the blood could have a number of non-skeletal health benefits.

The study, published online in PLOS ONE, reveals for the first time that improvement in the vitamin D status of healthy adults significantly impacts genes involved with a number of biologic pathways associated with cancer, cardiovascular disease (CVD), infectious diseases and autoimmune diseases. While previous studies have shown that vitamin D deficiency is associated with an increased risk for the aforementioned diseases, these results go a step further and provide direct evidence that improvement in vitamin D status plays a large role in improving immunity and lowering the risk for many diseases.

Vitamin D is unique in that it can be both ingested and synthesized by the body with sun exposure. It is then converted by both the liver and kidneys to a form that the body can use. An individuals' level of vitamin D, or their vitamin D status, is determined by measuring the level of 25-hydroxyvitamin D in the blood. Vitamin D deficiency, which is defined as a status of less than 20 nanograms per milliliter (ng/mL) of 25-hydroxyvitamin D, can cause a number of health issues, including rickets and other musculoskeletal diseases. Recently, however, data suggests that vitamin D deficiency (<20 ng/mL) and vitamin D insufficiency (between 21-29 ng/mL) is linked to cancer, autoimmune diseases, infectious diseases, type 2 diabetes and cardiovascular disease.

The randomized, double-blind, single-site pilot trial involved eight healthy men and women with an average age of 27 who were vitamin D deficient or insufficient at the start of the trial. Three participants received 400 International Units (IUs) of vitamin D per day and five received 2,000 IUs per day for a two-month period. Samples of white blood cells (immune cells) were collected at the beginning of the two-month period and again at the end. A broad gene expression analysis was conducted on these samples and more than 22,500 genes were investigated to see if their activity increased or decreased as a result of the vitamin D intake.

At the end of the pilot, the group that received 2000 IUs achieved a vitamin D status of 34 ng/mL, which is considered sufficient, while the group that received 400 IUs achieved an insufficient status of 25 ng/mL.

The results of the gene expression analysis indicated statistically significant alterations in the activity of 291 genes. Further analysis showed that the biologic functions associated with the 291 genes are related to 160 biologic pathways linked to cancer, autoimmune diseases, infectious diseases and CVD.

Examining gene response elements, or sequences of DNA bases that interact with vitamin D receptors to regulate gene expression, they also identified new genes related to vitamin D status. To ensure that their observations were accurate, the researchers looked at 12 genes whose level of expression does not change, and those genes remained stable throughout the trial period.

"This study reveals the molecular fingerprints that help explain the non-skeletal health benefits of vitamin D," said Michael F. Holick, PhD, MD, professor of medicine, physiology and biophysics at BUSM and leading vitamin D expert who served as the study's corresponding author. "While a larger study is necessary to confirm our observations, the data demonstrates that improving vitamin D status can have a dramatic effect on gene expression in our immune cells and may help explain the role of vitamin D in reducing the risk for CVD, cancer and other diseases."

A political activist was found dead in his home on March 12, the B.C. Coroners Service confirmed on Monday.

The coroner?s service is continuing to investigate what caused the death of John Boncore, 61, although foul play has been ruled out. (ahem!!!)

An initial account indicated that Boncore died after falling on a path on the Adams Lake reserve near Chase, but that was not the case, said Barb McLintock with the coroner?s office in Victoria.

?We have a very close description of where he was found and there is no question,? McLintock said, adding that someone may have seen him earlier on the trail. ?He was unequivocally in his house.?

Boncore?s friends in Alberta were concerned that his sudden death should be thoroughly investigated because his activism could have made him a target for violence. He was a carpenter and actor, but worked for decades on the front lines of activism, most recently in opposition to the Northern Gateway pipeline.

Larry McKillop, one of those friends, indicated that they hoped for an inquiry or an independent autopsy. He was told that neither one is a possibility, but that the coroner?s autopsy and toxicology results should be known in a month?s time, not four months as originally stated.

He felt it was premature to dismiss Boncore?s death as simply an accident or possibly due to alcohol. (smear campaign!)

?I?d hate for this to be swept under the rug if something had happened,? he said.

McKillop said he was told that Boncore fell on concrete in his bedroom, where his body was found. He is believed to have died the previous day.

?Somebody dropped him off on Monday night,? McKillop said. ?That was the last person who saw him. He was found dead on Wednesday morning.?
